Yes. The hardware and software of the ImmunoSpot® analyzers all support single- and double-color analysis. The optics and cameras of the S5 Micro Analyzers, however, are specifically enhanced for highest color fidelity and resolution.

fold faq What is the image resolution of the ImmunoSpot® analyzers?

Image resolution varies depending on the Analyzer model. The S5 Core and S5 Macro allow for a resolution of 1600 x 1200. With CTL SmartSegmentation, the S5 Micro can provide resolution of 4800 x 4800 pixels per well.  The S5 UV analyzer provides a resolution of 1388 x 1038 pixels per well.

ImmunoSpot® Analyzers use a proprietary light configuration to evenly illuminate the entire well bottom surface. In addition, the S5 model line features an auto-light adjustment option which permit fine-tuning of the illumination for each well when the background varies strongly.

fold faq Which types of assays might make high resolution necessary?

With spots in all size categories, high resolution images provide more information on the spot morphology, thereby increasing the accuracy of the advanced spot morphology recognition features offered by ImmunoSpot® software (SmartSpot™). The spot size reflects the per cell productivity and secretion kinetics. Also, the strength of T cell stimulation defines the spot size; low avidity T cell recognition results in low productivity and small, faint spots. Granzyme B, TNF and IL-10 assays frequently produce small spots. In all these cases, using high-resolution image analysis can provide a definite advantage.

fold faq Do the pictures include the entire well bottom, or will the rims be cut off?

The well images include the entire well bottom. Thanks to the auto-centering feature in the ImmunoSpot® analyzers, every single well is automatically centered using intelligent software algorithms. Thus, ImmunoSpot® analyzers compensate for manufacturing variations in the microtiter plates. Additionally, the ImmunoSpot® analyzers include Accurate Well Capture, a feature in which 100% of the well bottom surface is captured using autocentering with user definable safety margins.
